Inhibition of Superoxide Dismutase in Human Amnion, Chorion and Placenta by Cyanide and Azide

Abstract
Distribution of cytosolic and mitochondrial superoxide dismutase in human amnion, chorion and placenta and their inhibition by cysnide and azide were investigated. Activities of superoxide dismutase were 118.43¡¾3.29 U/g of chorion, 49.09¡¾5. 57 U/g of amnion and 31.55¡¾3.17 U/g of placenta. In amnion, predominsnt activity (93%) was found in cytosolic fraction complete inhibition of the activities of cytosolic superoxide dismutase in amnion, chorion and placenta were achieved by 5 mM, 10 mM and 6 mM cyanide, respectively. Mitochondrial superoxide dismutase from amnion and placenta were completely inactivated by 8 mM and 5 mM cyanide, respectively. However, only 40% of chorionic mitochondrial superoxide dismutase was inactivated by 10 mM cyanide. Inhibitory effect of azide was more remarkable to the cytosolic enzyme than to the mitochondrial enzyme.
